Western blot analysis of SIRT6 using anti-SIRT6 antibody (PB9357). The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human U87 whole cell lysates,
Lane 2: human A549 whole cell lysates,
Lane 3: human K562 whole cell lysates,
Lane 4: human U2OS whole cell lysates,
Lane 4: human Hela whole cell lysates.
After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-SIRT6 antigen affinity purified polyclonal antibody (PB9357) at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for SIRT6 at approximately 39-42 kDa. The expected band size for SIRT6 is at 39 kDa.

SIRT6 was detected in a paraffin-embedded section of mouse cardiac muscle tissue. Biotinylated goat anti-rabbit IgG was used as secondary antibody. The tissue section was incubated with rabbit anti-SIRT6 Antibody (PB9357) at a dilution of 1:200 and developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB (Catalog # AR1027) as the chromogen.

SIRT6 is also known as SIR2L6. This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class IV of the sirtuin family. Alternative splicing results in multiple transcript variants.
